  • We investigated the effect of aqueous extract of Cichorium intybus (CIAE) on mast cell-mediated immediate type allergic reactions. CIAE dose-dependently inhibited systemic anaphylactic reaction induced by compound 48/80 in mice. Especially, CIAE inhibited compound 48/80-induced anaphylactic reaction 100% with the dose of 1000 mg/kg. CIAE 1000 mg/kg also significantly inhibited local anaphylactic reaction activated by anti-dinitrophenyl (DNP) IgE. When mice were pretreated with CIAE at a concentration ranging from 0.1 to 1000 mg/kg, the plasma histamine levels were reduced in a dose-dependent manner. CIAE (1 to 1000 g/ml) dose-dependently inhibited histamine release from the rat peritoneal mast cells (RPMC) activated by compound 48/80 or anti-DNP IgE. These results indicate that CIAE inhibits mast cell-mediated immediate-type allergic reactions.

  • This report describes an inhibitory effect of Tongku-tang(TKT) on mast cell-mediated immediate-type allergic reactions. TKT is an Oriental herbal prescription, which has been successfully applied for the treatment of allergic disorders, mainly skin anaphylactic diseases in eastern medicine. TKT has concentration-dependently inhibited the ear swelling response induced by intradermal injection of non-specific mast cell degranulator compound 48/80 in mice. TKT also inhibited mast cell-dependent passive cutaneous anaphylaxis activated by dinitrophenyl (DNP)-IgE antibody in rats. I studied the effect of TKT on the histamine and β-hexosaminase release from the rat peritoneal mast cells by compound 48/80. TKT did not inhibit significantly the histamine and β-hexosaminase release from the rat peritoneal mast cells by compound 48/80. However, TKT inhibited both TNF-α and IL-1β secretion induced by phorbol 12-myristate 13-acetate and A23187 respectively. These results provide evidence that TKT may be beneficial in the treatment of immediate-type allergic reaction.

  • The effect of aqueous extract of Isodon japonicus Hara (Labiatae) (IJAE) on mast cell-mediated immediate-type allergic reactions was investigated. IJAE inhibited compound 48/80-induced systemic anaphylaxis and immunoglobulin E (IgE)-mediated local anaphylaxis. When IJAE was pretreated at the same concentration with systemic anaphylaxis, serum histamine levels were reduced in a dose-dependent manner. IJAE dose-dependently inhibited histamine release from rat peritoneal mast cells (RPMC) activated by compound 48/80. The level of cAMP in human mast cell line (HMC-1) cells, when IJAE was added, significantly was increased, compared with that of normal control. These results indicate that IJAE will beneficial in the treatment of immediate-type allergic reaction.

  • The mast cell plays a pivotal role in initiating allergic response by secreting intracytoplasmic granular mediators such as histamine. Magnoliae flos has been used for treatment of allergic disease in Korea. However, its effect in experimental models remains unknown. The present report describes an inhibitory effect of Magnoliae flos on mast cell-mediated immediate-type allergic reactions. Topical application of compound 48/80 can induce an ear swelling response in normal($WBB6F_1-+/+$) mice but not in the congenic mast cell-deficient $WBB6F_1-W/W^v$ mice. Magnoliae flos inhibited concentration-dependently mast cell-dependent ear swelling response induced by compound 48/80 by topical application. Magnoliae flos inhibited concentration-dependently passive cutaneous anaphylaxis induced by anti-dinitrophenyl (DNP) IgE in rats by topical application. Magnoliae flos also inhibited concentration-dependently the histamine release from the rat peritoneal mast cells by compound 48/80 or anti-DNP IgE. Moreover, Magnoliae flos had a significant inhibitory effect on compound 48/80-induced systemic anaphylactic reaction. These results indicate that Magnoliae flos inhibits immediate-type allergic reactions by inhibition of mast cell degranulation in vivo and in vitro.

  • The immediate-type allergic reaction (anaphylaxis) is involved in many allergic diseases such as asthma, allergic rhinitis, and sinusitis. The discovery of drugs for the treatment of immediate-type allergic diseases is a very important subject in human health. In this study, we investigated the effect of Rubus coreanus Miq.(Rosaceae) unripe fruits (RCF) on mast cell-mediated allergic reaction and inflammatory cytokine secretion. RCF inhibited compound 48/80-induced systemic reactions in mice. RCF attenuated immunoglobulin (Ig) E-mediated local allergic reactions. In addition, RCF dependently reduced histamine release from rat peritoneal mast cells local allergic reactions. In addition, RCF dependently reduced histamine release from rat peritoneal mast cells activated by compound 48/80 or IgE. Furthermore, RCF decreased the phorbol 12-myristate 13-acetate plus calcium ionophore A23187-stimulated tumor necrosis factor $(TNF)-{\alpha}$ and interleukin (IL)-6 secretion in human mast cells. Our findings provide evidence that RCF inhibits mast cell-derived immediate-type allergic reactions.

  • Immediate-type hypersensitivity is involved in many allergic diseases such as asthma, allergic rhinitis and anaphylaxis. The discovery of drugs for the treatment of allergic disease is an important subject in human health. Stimulation of mast cells releases inflammatory mediators, such as histamine and pro-inflammatory cytokines with immune regulatory properties. We investigated the effect of the aqueous extract of Schizonepeta tenuifolia (AEST) (Labiatae) on the immediate-type allergic reaction. AEST inhibited compound 48/80-induced systemic allergic reaction. AEST attenuated immunoglobulin E (IgE)-mediated skin allergic reaction and histamine release from human mast cell line (HMC-1) cells. In addition, AEST decreased the gene expression and secretion of pro-inflammatory cytokines in phorbol 12-myristate 13-acetate (PMA) plus calcium ionophore A23187 (A23187)-stimulated HMC-1 cells. Our results indicate that AEST inhibits the mast cell-derived allergic reactions and involvement of histamine and pro-inflammatory cytokines in these effects.

  • The effect of aqueous extract of Lycopus lucidus Turcz. (Labiatae)(LLAE) on mast cell-mediated immediate-type allergic reactions was investigated. LLAE (0.01 to 1 mg/g) dose-dependently inhibited systemic anaphylaxis induced by compound 48/80. LLAE (0.001 to 1 mg/g) also dose-dependently inhibited local anaphylaxis activated by anti-dinitrophenyl (DNP) IgE. When LLAE was pretreated at the same concentration with systemic anaphylaxis, serum histamine levels were reduced in a dose-dependent manner. LLAE (0.001 to 1 mg/mι) dose-dependently inhibited histamine release from rat peritoneal mast cells (RPMC) activated by compound 48/80. • Mahwang-Shin-Gung-San has been found inhibiting the mast cell-mediated anaphylactic reaction. This report describes an inhibitory effect of Mahwang-Shin-Gung-San (MSGS) on the immediate-type cutaneous allergic reactions. MSGS has concentration -dependently inhibited the ear swelling response induced by compound 48/80 in mouse by intradermal injection. The mast cells in mouse ear tissue undergone ear-swelling response by compound 48/80 were stained by alcian blue/nuclear fast red. MSGS significantly inhibited the compound 48/80-induced degranulation from mast cells in ear tissue. MSGS concentration-dependently inhibited the histamine release from the rat peritoneal mast cells (RPMC) by compound 48/80. We also studied the effect of MSGS on mast cell-dependent passive cutaneous anaphylaxis (PCA) activated by dinitrophenyl IgE antibody. MSGS showed potent inhibition of PCA by oral administration. These results indicate that MSGS inhibits immediate-type allergic reactions by inhibition of mast cell degranulation in vivo and in vitro.
