The Effect of Schizandrae Fructus Extract on Alcohol Fermentation and Enzyme Activities of Saccharomyce cerevisiae (오미자(Schizandra chinensis B.) 추출물이 Saccharomyces cerevisiae의 알콜발효 및 효소 활성에 미치는 영향)

  • The effect of Schizandrae fructus extract on the physiological properties of Saccharomyces cerevisiae was investigated. S. cerevisiae was inoculated into glucose broth, added with Schizandrae fructus extract, 0, 0.01, 0.1, 0.5 and 1%(w/v), respectively. And a 96 hours incubation was followed to investigate the changes in the growth, alcohol production, alcohol dehydrogenase and pyruvate decarboxylase activities of S. cerevisiae. The growth of S. cerevisiae was more pronounced in the broth containing 0.1 and 0.OlfS Schizandrae fructus extract than in the control. The growth was, however, inhibited in the broth containing 0.5 and 1% of the extract. The content of alcohol produced by S. cerevisiae also showed very similiar results with those of the yeast growth by addition of Schizandrae fructus extract. Alcohol dehydrogenase activities of S. cerevisiae cultured in broth treated with the extract of 0.1% and 0.01% increased by 25% and 18% than those in control group. Pyruvate decarboxylase activities in 0.1% and 0.01% treatments increased to 1.32 and 1.26 times. The activities in 0.5% and 1% treatments, however, decreased by 30% and 44%.

The Effects of Schizandrae Fructus Chloroform Fraction on Gene Expression in Liver Tissue of Dyslipidemic Mice (오미자(五味子) 클로로포름 분획물이 이상지질혈증 생쥐의 지질대사 및 간 조직 유전자 변화에 미치는 영향)

  • Objectives: Schizandrae fructus (Schizandra chinensis) is one of very common herbs, it is known as natural antioxidants, anti-inflammatory agent. Also some reports show that its extract works to regulate of dyslipidemia. This study was designed to investigate the effects of Schizandrae fructus chloroform fraction (SFCF) on serum lipid levels in dyslipidemic mice. Methods: The levels of total cholesterol, high density lipoprotein-cholesterol, triglyceride, aspartate aminotransferase (AST), alanine aminotransferase (ALT), fasting blood glucose in serum were measured. Histopathological and gene expression changes in liver tissue were also observed. Results: Oral administration of SFCF lowered levels of total cholesterol and triglyceride, which were elevated by high-fat diet. But SFCF did not affect on weight changes and serum AST, ALT levels in dyslipidemic mice. After carrying out gene ontological analysis, large numbers of genes in high-fat diet group were up-(347) or down-regulated (235). In SFCF treated mice, some changed expression of the genes was restored to normal levels, with a recovery rate of 17%. And it seems that fatty acid biosynthesis pathway was one of important key pathways to recovery. Conclusions: SFCF has beneficial effect on dyslipidemia, and could be used to prevent and treat cardiovascular disease.

Synergic Effect of Methanol extracts of Schizandrae Fructus and Mume Fructus on Experimental Mouse Colitis Induced by Dextran Sulfate Sodium (Dextran Sulfate Sodium 유도 마우스 대장염에 미치는 오미자와 매실의 상승효과)

  • The fruits of Schisandra chinensis and Prunus mume have been traditionally used in the Oriental countries as an astringent against diarrhea and abdominal pain, a protectant for liver disease, an antimicrobial, and a blood tonic. However, little is known about the extract of Schizandrae Fructus and Mume Fructus (SMF-Ex) on dextran-sulfate sodium (DSS)-induced colitis in mice. In this study, we investigated the protective effects of SMF-Ex on DSS-induced colitis in mice. An experimental colitis was induced by daily treatment with 5% DSS. SMF-Ex was orally administrated the single dose (80 mg/kg, body weight/day) for 7 days with one time per day. SMF-Ex reduced significantly clinical sign of DSS-induced colitis, including body weight loss, shorten colon length, increased disease activity index (DAI), and histological colon injury. SMF-Ex also inhibited significantly nitric oxide (NO) and prostaglandine $E_2$ ($PGE_2$) productions in DSS-induced colitis mice. Furthermore, SMF-Ex increased significantly an superoxide anion (SOD), catalase, and glutathione peroxidase (Gpx) activity of the colon tissue in DSS-induced colitis mice. These results suggest that SMF-Ex administration could reduce significantly the clinical signs and inflammatory mediators, and increase antioxidant activity in DSS-induced colitis model mice and is a good candidate for further evaluation as an effective anti-ulcerative agent.

Volatile components of Schizandra Chinensis Bullion (오미자(Schizandra Chinensis Bullion)의 휘발성 성분)

  • The essential oil of Schizandrae Fructus was extracted by simultanous distillation-extraction and steam distillation and analyzed by GC and GC-MS. The oil content of Schizandrae Frutus was 0.64% when extracted by steam distillation and 0.88% in case of simultanous distillation extraction and sensory analysis of the oil indicated woody, camphorous, spicy and sour characteristic notes. The experimental results confirmed the presence 47 volatile components, the major components were ${\gamma}-terpinene$, p-cymene, ${\alpha}-ylangne$ and ${\beta}-elemene$. The oil was fractioned into ten fractions and 4 fraction of them have a good aroma character. The application of the oil showed the utilization possibility as flavoring materials for Korea tea.

  • In the course of search for hepatoprotective agents from oriental traditional medicines, one compound was isolated from the MeOH extract of the fruits of Schizandra chinensis, and its structure was determined as gomisin J(1). The evaluation for hepatoprotective activity of isolated compound 1 on drug-induced cytotoxicity was conducted, and compound 1 showed protective effect with an EC$_{50}$value of 86.0 ${\pm}$ 5.3 ${\mu}$M against tacrine-induced cytotoxicity in Hep G2 cells. One of the well-known hepatoprotective agents, silybin, used as a positive control, and the hepatoprotective effect of compound 1 is similar with that of silybin (EC$_{50}$ value = 90.5 ${\pm}$ 9.8 ${\mu}$M).

Effect of Schisandrae Chinensis Fructus on Keratinocyte Damage by UV Irradiation (오미자(五味子)가 자외선유발 피부각질세포 손상에 미치는 효과)

  • This study was performed to assess the Effect of SCF(Schisandrae Chinensis Fructus) on Keratinocyte Damage by UV irradiation. The effect of SCF were determined in UV irradiated HaCaT. We measured LDH release and NO release from HaCaT to elucidate the effect of SCF. And iNOS, TNF-${\alpha}$, COX-2, Bax, Bcl-2, Bcl-xL, c-jun, c-fos gene expression were determined in HaCat using real time PCR method. The results are as follows. SCF inhibited LDH-release, NO production in UV irradiated HaCaT. SCF increased the gene expression Bax, Bcl-2 and Bcl-xL protein in UV irradiated HaCaT. SCF suppressed the gene expression TNF-${\alpha}$ in UV irradiated HaCaT. SCF suppressed the gene expression iNOS, c-fos, and c-jun in UV irradiated HaCaT. SCF not affected the suppression of the gene expression COX-2 in UV irradiated HaCaT. The study showed SCF inhibited the cell damage in UV irradiated HaCaT.

Antioxidant Activity and Inhibition of MMP-1 Expression of Schizandrae fructus (Schizandra chinensis) Extract (오미자 추출물의 항산화 활성 및 MMP-1 발현저해)

  • We invesitagat the effects of antioxidant and suppression of MMP-1 in UV-irradiated human dermal fibroblasts (HDF) by schizandrin isolated from Schisandra chinensis. Ultrasonic technique was used to extract schizandrin from S. chinensis. The higher contents of schizandrin (6.34 mg/ml) in 70% EtOH extract obtained by ultrasonic treatment at 20 MHz for 2 h during 10 days. Schizandrin had antioxidant activities. Schizandrin (1-10 ${\mu}g/ml$) induced a significant dose-dependent decrease in the expression fo MMP-1 protein. These results suggest that schizandrin is a potential candidate for the prevention and treatment of skin aging.

Screening of Antimicrobial Activity for Medicinal Herb Extracts (한약재 추출물의 항균효과 검색)

  • Twenty kinds of medicinal herbs were extracted by water and 95% ethanol and then antimicrobial activity of the extracts was investigated against various kinds of microorganisms. Water extracts of Gardeniae fructus (Gardenia jasminoides), Lycii fructus (Lycium chinense) and Schizandrae fructus (Schizandra chinensis) showed inhibitory effects on the growth of most of the bacteria. In the case of ethanol extracts, the 3 kinds of the samples such as Gardeniae fructus, Schizandrae fructus and Lithospermi radix (Lithospermum erythrorhizon) showed inhibitory effects on the growth of almost all bacteria. In particular, ethanol extract from Phellodendri cortex (Phellodendron amurense) showed the best inhibitory effect on the growth of S. aureus in the concentration of 0.01%. By the way, inhibitory effects of water extracts from these medicinal herbs were not so good on the growth of fungi but those of ethanol extracts were better and ethanol extracts of Phellodendri cortex showed best. Antimicrobial activity was variable according to the used extracting solvent. For example, inhibitory effets of ethanol ext-racts were $2{\sim}100$ times better than those of water extracts. Ethanol extract of Lithospermi radix was the most effective not only bactericidal effects but also sensory evaluation tests for tastes.
