Inhibitory Effect of Schizandrin on Nephrotoxicity of Cisplatin (오미자성분 Schizandrin의 Cisplatin유도 신장 독성에 대한 억제효과)

  • Cisplatin is one of the most effective antitumor agents currently available for cancer chemotherapy. However its clinical use has been limited by its severe side effects, especially nephrotoxicity. To evaluate the effect of schizandrin, one of radical scavengers and constituents of Schizandra chimensis, cisplatin and schizandrin were given intraperitoneally. Protective effect of schizandrin against nephrotoxicity of cisplatin was observed when schizandrin was administerd to rats 1,24 hr after cisplatin injection. Hepatotoxicity induced by combination treatment of cisplatin and schizandrin was evaluated by measuring sGPT and sGOT. Combination treatment did not affect the levels of sGPT and sGOT. The present result indicate that schizandrin when it is given after cisplatin, may provide protection against cisplatin nephrotoxicity without hepatotoxicity.

Schizandrin, Oil Compounds, and Their Extraction Yield in Fruits of Schizandra chinensis Baillon (오미자의 Schizandrin과 기름성분 및 추출수율 변화)

  • Histochemical distribution and varietal variation of schizandrin and oil compounds, and the changes of their extraction yield were investigated in fruits of collected Schizandra chinensis including Inje cultivar. In histochemical analysis on the distribution of schizandrin and oil in fruits of Inje cultivar, higher concentrations of them were found in the seed (1.01% and 27.6%, respectively) than in the epicarp and mesocarp of the whole fruit. Average contents of schizandrin in fruits and oil in seeds of collected lines were 0.84% and 27.9%, respectively. The mean composition of fatty acids in seeds oil was 3.6% of palmitic acid, 0.6% of stearic acid, 19.7% of oleic acid, 73.0% of linoleic acid, and 3.1% of linolenic acid, showing high composition(95.8%) of total unsaturated fatty acid. Oil extracted from seeds of Inje cultivar contained 4.29% of schizandrin, indicating that seed oil contained much schizandrin, a bioactive lipid-soluble compound. Compared with 80% methanol extraction in fruits and seeds, yields of schizandrin and oil were lower showing 23.8% and 17.3%, respectively in boiling water extraction of the fruits and seeds without grinding. The seeds soaked with water during four months contained 1.18% of schizandrin and 25.2% of oil, whose contents were similar to those of the seeds stored at room temperature. These results demonstrated that the seed in the whole fruit could be utilized as a source to extract its functional oil and bioactive lipid-soluble compounds like schizandrin, especially after using Schizandra fruits for the beverage manufacture.

  • Omija (Schizandra chinensis Baillon) has five lignans such as schizandrin, gomisin A, gomisin N, gomisin C and schizandrin C. Its major constituent, schizandrin, has a deterrent effect on nephrotoxicity of cisplatin, blood sugar and an ulcer of the stomach. In this study, we have investigated optimized extraction condition to make high functional beverage by quantifying schizandrin and measuring of browning index. Browning index was used as an index of color deterioration of anthocyanin. Browning index was decreased according to increment in extraction temperature and ethanol concentration. The optimum extraction time, extraction temperature and alcohol concentration levels were 3 hrs, $46^{\circ}C$ and 24% alcohol for high schizandrin concentration and desired browing index.

  • Schizandrae Fructus has been used for controlling respiratory allergic or inflammatory diseases in folk medicine and their components, schizandrin, schizandrin-A and gomisin-A were reported to have diverse biological effects. In this study, we investigated whether schizandrin, schizandrin-A and gomisin-A affect adenosine triphosphate (ATP)-induced mucin secretion from cultured airway epithelial cells. Confluent primary hamster tracheal surface epithelial (HTSE) cells were metabolically radio labeled using $^{3}H-glucosamine$ for 24 h and chased for 30 min in the presence of varying concentrations of each agent to assess the effects on $^{3}H-mucin$ secretion. The results were as follows: 1) schizandrin significantly inhibited ATP-induced mucin secretion; 2) However, schizandrin-A and gomisin-A did not affect ATP-induced mucin secretion, significantly. We conclude that schizandrin can inhibit ATP-induced mucin secretion by directly acting on airway mucin-secreting cells. Therefore, schizandrin should further be investigated for the possible use as mucoregulators in the treatment of inflammatory airway diseases.

Antioxidant Activity and Inhibition of MMP-1 Expression of Schizandrae fructus (Schizandra chinensis) Extract (오미자 추출물의 항산화 활성 및 MMP-1 발현저해)

  • We invesitagat the effects of antioxidant and suppression of MMP-1 in UV-irradiated human dermal fibroblasts (HDF) by schizandrin isolated from Schisandra chinensis. Ultrasonic technique was used to extract schizandrin from S. chinensis. The higher contents of schizandrin (6.34 mg/ml) in 70% EtOH extract obtained by ultrasonic treatment at 20 MHz for 2 h during 10 days. Schizandrin had antioxidant activities. Schizandrin (1-10 ${\mu}g/ml$) induced a significant dose-dependent decrease in the expression fo MMP-1 protein. These results suggest that schizandrin is a potential candidate for the prevention and treatment of skin aging.

Antitumor Effect of Schizandrin by Inhibiting Angiogenesis (Schizandrin의 신혈관형성억제에 의한 항암효과)

  • Schizandra chinensis extract has been known to possess a variety of efficacy including antitumor. However, it remains unclear how schizandrin, which is a major biological active ingredient of Schizandra chinensis, exerts antitumor effect. This study was designed to investigate the mechanism by which schizandrin inhibits tumor growth and metastasis. In in vivo test using tumor model mice injected with B16BL6 cell line, mice treated with 10 and 100 ${\mu}g/ml$ schizandrin showed a significant inhibition by $73.79{\pm}6.43%$ and $90.46{\pm}1.72%$, respectively, compared with positive tumor controls. Schizandrin did not exert a significant toxicity for the normal cells (HUVECs) and tumor cell lines (A549, B16BL6, Du145, Huh7). Treatment with schizandrin at 10 and 100 ${\mu}g$/head significantly inhibited the tumor-induced angiogenesis by $68.04{\pm}32.21%$ and $103.8{\pm}34.99%$ compared with the positive control group, respectively. Using in vivo lung metastasis model, tumor metastasis assay revealed that 10 and 100 ${\mu}g$/head schizandrin significantly decreased the metastatic lung tumor by $37.51{\pm}8.15%$ and $75.53{\pm}4.38%$ compared with positive controls, respectively. On the other hand, schizandrin did not affect the adherence of B16BL6 cell line to extracellular matrix protein. These results demonstrate that schizandrin exerts inhibitory effect on tumor growth and metastasis by inhibiting angiogenesis. This study thus suggest that schizandrin may be a candidate molecule target for cancer drug development.

Confirmation of Schizandrin as a Marker Compound in Jangsu Omija Powder (HPLC를 이용한 기능성 원료 지표성분으로서 장수 오미자 분말 중 Schizandrin의 분석법 개발)

  • We studied the development of schizandrin as a marker compound in Jangsu Omija. Schizandrin was validated for its LOD (limit of detection), LOQ (limit of quantitation), precision, accuracy, and recovery by HPLC relative to Omija powder. It showed a high linearity in the calibration curve with a coefficient of correlation ($R^2$) of 0.9990. The LOD and LOQ were 0.2 ${\mu}g/mL$ and 0.5 ${\mu}g/mL$, respectively. The intra- and inter-day precision of schizandrin calibration standards was 0.06~0.66% and 0.13~1.19%, respectively, and the intra- and inter-day accuracy of schizandrin was 98.35~103.30% and 98.35~103.00%, respectively. Overall, schizandrin was validated through our analytical methods as a marker compound in Jangsu Omija.

Effects of Baicalin, Baicalein and Schizandrin on Airway Mucin Production Induced by Epidermal Growth Factor and Phorbol Ester

  • We conducted this study to investigate whether baicalin, baicalein or schizandrin significantly affect MUC5AC mucin production induced by epidermal growth factor (EGF) or phorbol ester (PMA) in human airway epithelial cells. Confluent NCI-H292 cells were pretreated with varying concentrations of baicalin, baicalein or schizandrin for 30 min and then stimulated with EGF or PMA for 24 h, respectively. MUC5AC mucin protein production was measured by ELISA. The results were as follows: (1) Baicalin was found to inhibit the production of MUC5AC mucin protein induced by both EGF and PMA. (2) Baicalein, the aglycone of baicalin, also inhibited MUC5AC mucin production. (3) Schizandrin, derived from Schizandrae Fructus, inhibited MUC5AC mucin production by the same inducers. These results suggest that baicalin, baicalein and schizandrin can regulate the production of mucin protein by directly acting on human airway epithelial cells.

Anti-proliferative and Pro-apoptotic Activities by Pomace of Schisandra chinensis (Turcz.) Baill. and Schizandrin (오미자 박 추출물 및 schizandrin에 의한 암세포 항성장 및 세포사멸 활성)

  • Schisandra chinensis (Turcz.) Baill. (omija) is often used in Chinese medicine to treat various human diseases, and is known to possess various bioactive components such as schizandrin and gomisin A. In the present study, we prepared ethanol extracts of pomace of Schisandra chinensis (PSC) and investigated their effects on cell viability and expression changes of pro-apoptotic genes such as ATF3, NAG-1 and p21 in human colorectal cancer HCT116 cells. PSC significantly reduced cell viability in a dose-dependent manner, and also dramatically induced the expression of ATF3, NAG-1 and p21 genes, with resveratrol used as a positive control. We also assessed the effects of pure compound schizandrin (SZ) derived from Schisandra chinensis on cell viability and expression of pro-apoptotic genes such as ATF3, NAG-1 and p21. The results showed that SZ also decreased cell viabilities in a dose-dependent manner and increased the expression of ATF3, NAG-1 and p21 genes. In addition, apoptosis was detected in SZ-treated HCT116 cells, which was confirmed with PARP cleavage. PARP cleavage was recovered in part by the transfection of NAG-1 siRNA. The results indicate that NAG-1 is one of the genes responsible for apoptosis induced by SZ. Overall, our findings may contribute to understanding the molecular mechanisms of anti-proliferative and pro-apoptotic activities mediated by PSC and SZ.

Quantitative Analysis of Lignans from Fruits of Schizandra chinensis (오미자 리그난 성분의 정량분석)

  • This study was carried out to establish the quantitative analysis of lignans for the quality evaluation of fruit from Schizandra chinensis. Five lignans, gomisin N, schizandrin, gomisin C, schisantherin C, and gomisin A were isolated from the fruits of S. chinensis and identified by the spectroscopic methods. Quantitative determination of three lignans, schizandrin, gomisin A, gomisin N, was conducted using HPLC. Average contents of three lignans in collected lines were 0.70% of schizandrin, 0.20% of gomisin A, 0.57% of gomisin N, and 1.47% of total lignans.
