Anti-inflammatory Effect of Patrinia villosa Extract on Proteinase-activated Receptor-2 Mediated Paw Edema (Proteinase 활성수용체-2 유발 흰쥐 발바닥 부종에 미치는 패장근 물추출물의 항염증 효과)

  • The root of Patrinia villosa Jussieu (Valerianaceae) has long been used for treatment of infectious diseases in Korea. In this study, the anti-inflammatory effect of the Patrinia villosa root water extract (PVWX) was investigated in proteinase-activated receptor-2 (PAR2)-mediated rat paw edema. Paw edema was induced by injection of trypsin or $trans-cinnamoyl-LIGRLO-NH_2\;(tc-NH_2)$ into hindpaw of rats. PVWX. (10, 50, 100 and 200 mg/kg) was orally administered 1 h before the induction of inflammation. At doses of 50, 100 and 200 mg/kg, PVWX. showed significant inhibition on both change in paw volume and vascular permeability. PVWX. (100 mg/kg) significant1y inhibited PAR2 agonists-induced myeloperoxidase (MPO) activity in paw tissue. These results indicate that PVWX has an anti-inflammatory action in PAR2-mediated paw edema.

  • Potentilla chinensis Ser. (Rosaceae) has long been used for a remedy of diarrhea and inflammation in Korea. In this study, the anti-inflammatory effects of the Potentillae chinensis Herba water extract (PCX) was investigated in proteinase-activated receptor-2 (PAR2)-mediated rat paw edema. Paw edema was induced by injection of trypsin or trans-cinnamoyl-LIGRLO-$NH_2$ (tc-$NH_2$) into the hind paw of rats. PCX (10, 50, 100 and 200 mg/kg) was orally administered 1 h before the induction of inflammation. At doses of 50, 100 and 200 mg/kg, PCX showed significant inhibition on both change in paw volume and vascular permeability. PCX (100 mg/kg) significantly inhibited PAR2 agonists-induced myeloperoxidase (MPO) activity in paw tissue. These results indicate that PCX has an anti-inflammatory action in PAR2-mediated paw edema.

Labeling IgG with $^{99m}Tc$ using 2-iminothiolane (2-iminothiolane을 이용한 IgG의 $^{99m}Tc$ 표지)

  • 2-iminothiolane is known to bind $NH_2$ group of lysine in the protein and deliver SH group, which can be used to label protein with $^{99m}Tc$. In this study, we looked for the best reaction condition in which 2-iminothiolane is conjugated to human polyclonal IgG and labeling condition with $^{99m}Tc$-glucoheptonate. Labeling yield was measured with TSK G4000SW column and HPLC or precipitation with 10% TCA (trichloroacetic acid) and 1% HSA. In vivo distribution was investigated with Staphylococcal abscess bearing rats. With decreasing glucoheptonate, the labeling yield decreased. Without 2-iminothiolane, $^{99m}Tc$-glucoheptonate was bound to IgG, which seemed to be direct labeling. With increasing 2-iminothiolane upto 20 times higher than IgG, the labeling yield increased, and plateau was seen with higher molar excess of 2-iminothiolane. Polymer formation was not observed. The pH for the conjugation of 2-iminothiolane and IgG was best around 6.4. $^{99m}Tc$-2-iminothiolane-IgG showed faster blood clearance, higher renal activity and lower hepatic and splenic activity than $^{99m}Tc$-DTPA-IgG. The biodistribution of $^{99m}Tc$-2-iminothiolane-IgG with higher molar excess of 2-iminothiolane was not different from that with lower molar excess. Labeling antibodies with $^{99m}Tc$ using 2-iminothiolane can afford a possible route to simple labeling and wide clinical use of the immunoscintigraphy.

Effects of Surface Nitrification on Thermal Conductivity of Modified Aluminum Oxide Nanofibers-Reinforced Epoxy Matrix Nanocomposites

  • Aluminum oxide ($Al_2O_3$) nanofibers were treated thermally under an ammonia ($NH_3$) gas stream balanced by nitrogen to form a thin aluminum nitride (AlN) layer on the nanofibers, resulting in the enhancement of thermal conductivity of $Al_2O_3$/epoxy nanocomposites. The micro-structural and morphological properties of the $NH_3$-assisted thermally-treated $Al_2O_3$ nanofibers were characterized by X-ray diffraction (XRD) and atomic force microscopy (AEM), respectively. The surface characteristics and pore structures were observed by X-ray photoelectron spectroscopy (XPS), Zeta-potential and $N_2$/77 K isothermal adsorptions. From the results, the formation of AlN on $Al_2O_3$ nanofibers was confirmed by XRD and XPS. The thermal conductivity (TC) of the modified $Al_2O_3$ nanofibers/epoxy composites increased with increasing treated temperatures. On the other hand, the severely treated $Al_2O_3$/epoxy composites showed a decrease in TC, resulting from a decrease in the probability of heat-transfer networks between the filler and matrix in this system due to the aggregation of nanofiber fillers.

  • The flower of Magnolia denudata Desrousseaux(Magnoliaceae) has long been used for treatment of nasal disorder in Korea. The physiological activity of the Magnoliae Flos ethanol extract (MFX) was investigated. MFX showed antimicrobial activity. At doses of 100 and 200 mg/kg, MFX showed significant inhibition on both change in paw volume and vascular permeability. MFX (100 mg/kg) significantly inhibited PAR2 agonist-induced myeloperoxidase (MPO) activity in paw tissue. These results indicate that MFX has anti-inflammatory activity in PAR2-mediated paw edema.

Adsorption and electro-Fenton processes over FeZSM-5 nano-zeolite for tetracycline removal from wastewater

  • Adsorption and heterogeneous electro-Fenton process using iron-loaded ZSM-5 nano-zeolite were investigated for the removal of Tetracycline (TC) from wastewater. The nano-zeolite was synthesized hydrothermally and modified through impregnation. The zeolite was characterized by XRD, FT-IR, FE-SEM, N2 adsorption-desorption, and NH3-TPD techniques. The equilibrium data were best represented by the Freundlich isotherm. The pseudo-second-order kinetic model was the most accurate model for the adsorption of TC on the modified nano-zeolite. The effect of parameters such as pH of solution and current density were investigated for the heterogeneous electro-Fenton process. The results showed that the current density of 150 mA and pH of 3 led to the highest TC removal (90.35%) at 50 min. The nano-zeolite showed the appropriate reusability. Furthermore, the developed kinetic model was in good agreement with the removal data of TC through the electro-Fenton process.

Preparation of Nanoflake Bi2MoO6 Photocatalyst Using CO(NH2)2 as Structure Orientation and Its Visible Light Degradation of Tetracycline Hydrochloride

  • Bi2MoO6 (BMO) via the structure-directing role of CO(NH2)2 is successfully prepared via a facile solvothermal route. The structure, morphology, and photocatalytic performance of the nanoflake BMO are characterized by X-ray diffraction (XRD), scanning electron microscopy (SEM), fluorescence spectrum analysis (PL), UV-vis spectroscopy (UV-vis) and electrochemical test. SEM images show that the size of nanoflake BMO is about 50 ~ 200 nm. PL and electrochemical analysis show that the nanoflake BMO has a lower recombination rate of photogenerated carriers than particle BMO. The photocatalytic degradation of tetracycline hydrochloride (TC) by nanoflake BMO under visible light is investigated. The results show that the nanoflake BMO-3 has the highest degradation efficiency under visible light, and the degradation efficiency reached 75 % within 120 min, attributed to the unique hierarchical structure, efficient carrier separation and sufficient free radicals to generate active center synergies. The photocatalytic reaction mechanism of TC degradation on the nanoflake BMO is proposed.

Inhibitory Effect of Mouse Paw Edema by Solanum melongena Aqueous Extract (노랑가지 물추출물에 의한 생쥐 발바닥 부종의 억제효과)

  • Solanum melongena L. (Solanaceae) has anti-oxidant, analgesic, and hypolipidemic effects. We previously showed that Solanum melongena (SM) aqueous extract inhibits mast cell-mediated allergic reactions. The activation of protease-activated receptor-2 (PAR-2) induces acute inflammation in rat hindpaw. In the present study, we investigated the effects of the SM aqueous extract on mouse paw edema induced by PAR2 agonists. Trypsin or trans-cinnamoyl-LIGRLO-NH$_2$ (tc-NH$_2$), PAR-2 agonists, was injected into the hind paw of mice to induce paw edema. SM aqueous extract (1, 5, 10, and 100 mg/kg) was orally administered 1 hr before induction of paw edema. SM aqueous extract (5, 10, and 100 mg/kg) significantly inhibited both paw edema and vascular permeability in the dose-dependent manner. Furthermore, SM aqueous extract (10 mg/kg) significantly inhibited PAR-2 agonist-induced myeloperoxidase (MPO) activity and tumor necrosis factor (TNF)-$\alpha$ expression in paw tissue compared to that of saline. These results suggest that SM aqueous extract may be useful for treatment of PAR-2-mediated inflammation.

The Correlation between Acholic Stool and the Result of $Tc^{99m}$ DISIDA Hepatobiliary Scintigraphy and Biochemical Test in Neonatal Cholestasis (신생아 담즙 정체증에서 무담즙변의 유무와 $Tc^{99m}$ DISIDA 간담도 주사 결과간의 상관성과 생화학적 검사의 차이에 관한 연구)

  • Purpose: The most common causes of neonatal cholestasis are neonatal hepatitis (NH) and extrahepatic biliary atresia (EHBA). Since neonatal cholestasis presents with variable expression of same pathologic process and has similar clinical, biochemical, and histologic features between EHBA and idiopathic neonatal hepatitis (NH), differential diagnosis is often difficult. We reviewed the differences of clinical characteristics and laboratory data to find out any correlation between the results of $Tc^{99m}$ DISIDA scan and presence of acholic stool. Methods: Between June 1993 and January 2001, total 29 infants younger than 4 month-old underwent $Tc^{99m}$ DISIDA scan. Their biochemical tests and clinical course were reviewed retrospectively. Results: Patients who had negative intestinal activity on $Tc^{99m}$ DISIDA scan showed acholic stool and revealed higher serum direct bilirubin and urine bilirubin level. 18.2% of patients with acholic stool showed intestinal activity on $Tc^{99m}$ DISIDA scan and 81.8% of them did not. All the patients without acholic stool showed positive intestinal activity on $Tc^{99m}$ DISIDA scan. The result of $Tc^{99m}$ DISIDA scan and the presence of acholic stool showed high negative correlation (r :-0.858). Patients with acholic stool and negative intestinal activity on $Tc^{99m}$ DISIDA scan showed higher serum total bilirubin level. Patients without acholic stool and positive intestinal activity on $Tc^{99m}$ DISIDA scan showed higher serum level of ALT. Conclusion: Patients with acholic stool and negative intestinal activity showed high correlation, but 18.2% of patients with acholic stool showed positive intestinal activity. So operative cholangiogram or transcutaneous liver biopsy should be performed for confirmation.
