결장염 유발 Rat의 결장운동성에 침술이 미치는 영향 (Effects of Traditional Acupuncture on Colonic Motility in the Rat with Colitis)

  • 본 연구는 정상 및 결장염을 유발한 rat에서 침술이 결장운동성에 미치는 영향을 알아보고, 이에 내인성 opioid가 관여하는지의 여부를 알아보기 위하여 수행하였다. 22마리의 Sprague-Dawley rat을 정상군 (n=8), 결장염군 (n=6) 및 maloxone군(n=8)의 세군으로 나누어 실험을 실시하였다. Rat의 근위 결장의 장막하에 스테인레스 전극을 장착하고, 전극장착 7일 후에 trinitrobenzene sulphonic acid(TNBS)과 ethanol을 이용하여 결장염을 유발하였다. 결장의 근전도는 전극장착 11일 후에 trinitrobenzene sulphonic acid(TNBS)과 ethanol을 이용하여 결장염을 유발하였다. 결장의 근전도는 전극장착 11일 후에 polygraph를 이용하여 측정하였다. 정상군에서는 정상 결장의 기초 근전도를 60분간 기록한 후에 GV-1에 20분간 전통 침술을 실시하고, 다시 60분간 근전도를 가록하였으며, 결장염군에서는 TNBS/ethanol이 미리 투여된 rat에 정상군에서와 같은 방법으로 근전도 기록 및 침술을 실시하였다. Naloxone 군에서는 결장염군과 같은 방법으로 TNBS/ethanol이 미리 투여된 rat에 침술을 실시하기전 naloxone(3 mg/kg, SC)을 투여하고 다른 군에서와 마찬가지로 근전도와 침술을 실시하였다. 정상군에서는 침술이 결장의 운동성에 영향을 미치지 않았다. 그러나 TNBS/ethanol을 투여하여 결장염이 유발된 결장염군에서는 침술에 의해서 결장의 운동성이 유의적으로 저하되었다(p<0.01). Naloxone군에서는 침술에 의한 결장운동성의 저하는 관찰할 수 없었다. 이상의 결과로 보아 rat에서 GV-1 혈위의 침술에 의해 유발된 내인성 opioid는 염증상태의 결장 운동성을 감소시킨 것으로 사료된다.

합곡의 구진약침이 흰쥐의 염증성 대장염에 미치는 치료 효과 (Effects of Moxi-tar Herbal Acupuncture at LI4 (HapGok) on TNBS-induced Colitis in Rats)

  • Objectives : Ulcerative colitis or Crohn's disease has been recognized as Ha-ri (下痢) or Jang-Byok in Korean oriental medicine. A purpose of the present study is to investigate the anti-inflammatory effect of moxi-tar herbal acupuncture at LI4 (HapGok) on 2,4,6-trinitrobenzene sulphonic acid (TNBS) induced colitis in rats and further elucidate the possibility of herbal acupuncture on ulcerative colitis which is chronic inflammatory disease of the gastrointestinal tract. Methods : Sprague-Dawley rats, weighing $170{\sim}190$ g, were subjected to intrarectal injection of either saline (300 ${\mu}l$, 500 ${\mu}l$) for a control or 2,4,6-trinitrobenzene sulphonic acid (TNBS) (300 ${\mu}l$, 500 ${\mu}l$) for a colitis, Moxi-tar (20 mg/ml) were subcutaneously injected to the LI4 just after the secondary injection time of TNBS in rats. To study the effects of Moxi-tar acupuncture in LI4, body weight, RBC count, WBC count, total protein, Paw edema rate, rate of protein leakage into CMC-pouch fluid, IgG levels and IgM levels were observed. Results: Moxi-tar acupuncture in LI4 on TNBS-induced colitis inhibited the body weight lose rate but not effect RBC, WBC count. In addition, it inhibited the reduction of total protein concentration, paw edema, rate of protein leakage into CMC-pouch fluid, IgG levels and IgM levels. Conclusions : It is suggested that moxi-tar herbal acupuncture at LI4 helps to recover TNBS-induced colitis and plays an important role for an treatment of the irritable bowel syndrome (IBS).

TNBS 동물 모델과 loperamide 동물 모델에서 해삼 건조 분말의 대장염 및 변비 개선 효과 (Improving Effects of Stichopus Japonicus on TNBS-Induced Colitis and Loperamide-Induced Constipation in Animal Disease Models)

  • Colitis and constipation are the most common intestinal complaints worldwide. This study examined the beneficial effects of sea cucumber powder for TNBS-induced colitis in ICR mice(n=6/group) and loperamide-induced constipation in rats(n=8/group). Animals were divided into normal and four dried see cucumber treated groups that were named Nor, Con(0 mg/kg), GI(30 mg/kg), GII(100 mg/kg) and GIII(300 mg/kg). In order to induce colitis and constipation, 5% TNBS was injected into distal colon and loperamide (2 mg/kg, twice a day, peroral) was treated for 5 day in the four experimental groups but not the normal group. Gross finding score was decreased tendency by oral administration of sea cucumber in colitis-induced animal model, but colonic weight was not different. Excreted fecal pellet number, weight and water content were increased in the sea cucumber-treated group compared to the non-treated group. The fecal pellet number was reduced within distal colon of the sea cucumber-treated groups. Interestingly, intestinal transit length was increased compared to the non-treated group. Our results demonstrated that colitis and constipation were improved by sea cucumber treatment in the animal models. Specifically, gross finding score in TNBS-induced colitis model was improved by sea cucumber. After animals being fed sea cucumber, excreted fecal number, weight, water contents, and fecal number within distal colon and colonic transit length also get better in loperamide-induced constipation model.
